If V created his last Horcrux after murdering Frank Brice then
Wormtail would certainly know about it as he was there and V was
quite helpless (couldnt feed himself) without his assistance. Would
the very secretive V allow Wormtail to know one of his most guarded
secrets?

Do you really think Nagini can't take Wormtail?

I think it depends on how *fresh* the soul fragment needs to be. Frank
Bryce was murdered at the beginning of GoF. LV could have made Nagini a
Horcrux anytime after being resurrected in the graveyard without Wormtail
knowing about it.
Read exactly what DD says in HBP, Ch. 23:
"After an interval of some years, however, he used Nagini to kill an old
Muggle man, and it might then have occurred to him to turn her into his
last Horcrux."
He says LV got the idea after killing the old Muggle. He doesn't say that
LV actually used that murder to create a Nagini Horcrux. There were also a
few murders in OotP and HBP.

Why would Wormtail even what a Horcrux is? Voldemort could have
cast the spell right in front of him without him even realising,
especially if it was done non-verbally.
